Membrane proteins consisting of approximately 23% of the human proteome, play a crucial role in many physiological processes. Since most drugs achieve their therapeutic effects by interacting with membrane proteins, these proteins are also the major drug target category for pharmaceuticals (more than 60%). To date, there are some typical membrane protein targets, including GPCRs, transporters and ion channels. Over the past 20 years, new technologies and methods have greatly advanced the discovery and development of membrane protein-based drugs, including small molecule drugs and monoclonal antibody (mAb) therapeutics.

Membrane proteins as drug targets

According to their structure, membrane proteins can be divided into three types, including integral membrane proteins, peripheral membrane proteins, and lipid-anchored proteins. Among them, integral membrane proteins (including transmembrane proteins) anchored to membranes are important to drug targets across most fields of medicine. Increasing knowledge of functional modulators of integral membrane proteins, such as ligands, specific lipids/cholesterol, protein interaction partners, and nanobodies that stabilize desired conformations, can help select more specific integral membrane protein-targeting drugs in discovery analysis.
