Challenges in antibody discovery to complex membrane proteins
The diversity of the structures of membrane proteins and membrane-bound enzymes creates an abundance of opportunities for interventions that regulate the specific functions of these proteins. Moreover, this abundance also contributes to the promise of therapeutic antibodies against complex membrane proteins. The structures of antibodies can be engineered in many ways to match various targets. When analyzing families of possible drug target proteins, membrane-associated GPCRs dominate, followed by ion channels, transporter proteins, and membrane-bound enzymes. While these membrane proteins are a key group for therapeutic antibody applications, they also face a variety of challenges.

Membrane protein proteins are often expressed in low abundance and require significant time and effort to determine the optimal expression system.
- Membrane protein extraction and purification
When using membrane proteins as antigens, it is often necessary to isolate and extract the membrane protein. However, once the protein is removed from the lipid environment, it is usually unstable and the natural conformation of the protein needs to be stabilized.
- The structural and functional diversity of the membrane proteins
In addition, the structural diversity of integral membrane proteins is also challenged, by the fact that the required antibody epitopes may be small and inaccessible.
